Zinc and ring finger 1 (ZNRF1) is a membrane-associated RING-type E3 ubiquitin ligase primarily expressed in the nervous system, where it regulates neuronal signaling and axonal integrity. It contains a C3HC4 RING zinc finger domain that facilitates interaction with ubiquitin-conjugating (E2) enzymes to promote the transfer of ubiquitin to substrate proteins for proteasomal degradation. ZNRF1 is a key mediator of injury-induced axon degeneration, acting through the ubiquitination of signaling components involved in Wallerian-like processes. While its upregulation is known to accelerate axonal loss in experimental models of neurodegeneration and neuropathic injury, its role in human disease is currently characterized as modulatory.
