YES proto-oncogene 1, Src family tyrosine kinase (YES1) is a non-receptor tyrosine kinase belonging to the Src family of kinases (SFKs). It plays a pivotal role in promoting cell proliferation, survival, and invasiveness during tumor development. YES1 is amplified and overexpressed in many tumor types, including breast, lung, and ovarian cancers, where it is associated with poor prognosis and resistance to chemotherapeutic drugs and tyrosine kinase inhibitors. Structurally, YES1 contains myristoylation and palmitoylation sites in the SH4 domain, which anchor it to cell membranes, and key tyrosine phosphorylation sites (Y426 and Y537) that control its activation and inactivation. YES1 interacts with various signaling molecules, including YES-associated protein 1 (YAP1), to promote tumorigenesis.
