WNT inhibitory factor 1 (WIF1) is a secreted protein composed of 379 amino acids, featuring an N-terminal signal sequence, a unique and conserved 150-amino-acid WIF domain, five epidermal growth factor (EGF)-like repeats, and a hydrophilic C-terminal tail. WIF1 functions as a lipid-binding protein that directly interacts with a broad range of canonical and non-canonical Wnt ligands, such as Wnt3a, Wnt4, Wnt5a, Wnt7a, Wnt9a, and Wnt11, thereby inhibiting their ability to bind to membrane-bound receptors and suppressing Wnt/β-catenin signaling. Structural studies have confirmed its capacity to bind additional extracellular proteins, including olfactomedin 1 and connective tissue growth factor, suggesting a multifaceted regulatory role in extracellular signaling. In disease, WIF1 acts as a tumor suppressor, with its expression frequently downregulated in cancers such as osteosarcoma due to promoter hypermethylation; restoration of WIF1 expression in deficient tumor cells significantly reduces tumor growth and metastasis in vivo.
