Vanin 2 (VNN2) is a GPI-anchored cell surface amidohydrolase that exhibits potent pantetheinase activity. It functions by hydrolyzing D-pantetheine to recycle pantothenic acid (vitamin B5) and release cysteamine, a molecule involved in the cellular response to oxidative stress. Primarily expressed on myeloid cells within the hematopoietic system, VNN2 plays a crucial role in leukocyte dynamics, including adhesion, migration, and the transendothelial migration of neutrophils. It also regulates beta-2 integrin-mediated cell adhesion and is involved in the thymus homing of bone marrow-derived cells. Clinically, VNN2 is a well-established marker for B-cell precursor acute lymphoblastic leukemia (ALL). Its expression in ALL is associated with increased chemotherapy resistance, minimal residual disease, and a poorer clinical prognosis. Additionally, VNN2 serves as a marker for fetal liver hematopoietic stem cells, highlighting its importance in both normal hematopoiesis and leukemogenesis.
