Ubiquitin specific peptidase 7 (USP7) is a deubiquitinating enzyme that plays crucial roles in various cellular processes. It contains 1102 amino acids and consists of an N-terminal TRAF-like domain, a catalytic core domain, and five C-terminal ubiquitin-like domains. USP7 regulates protein stability by removing ubiquitin from target proteins, including p53, MDM2, and FOXO4. It is involved in DNA damage response, cell cycle regulation, and epigenetic modifications. USP7 also participates in immune signaling and maintenance of DNA methylation. Dysregulation of USP7 has been implicated in several diseases, particularly cancer. Aberrant expression and activity of USP7 are found in various cancers, contributing to tumor initiation and progression. Additionally, loss-of-function mutations in USP7 are associated with a neurodevelopmental disorder characterized by developmental delay, autism spectrum disorder, and epilepsy.
