Plasminogen activator, urokinase (Urokinase) is a secreted serine protease that converts plasminogen into active plasmin, thereby initiating fibrinolysis and extracellular matrix degradation. Synthesized as a single-chain zymogen, it is activated by plasmin into a two-chain form that binds the GPI-anchored urokinase receptor (uPAR). This interaction localizes proteolytic activity to the cell surface, modulating cell adhesion and migration, particularly at the tumor-stroma interface. Urokinase is composed of a growth factor domain, a kringle domain, and a catalytic serine protease domain. Clinically, urokinase is an FDA-approved thrombolytic agent used for the treatment of pulmonary embolism and other acute thrombotic disorders, where it effectively promotes the dissolution of fibrin clots.
