Ubiquitin conjugating enzyme E2 F (putative) (UBE2F) is a specialized NEDD8-conjugating enzyme that plays a critical role in the protein neddylation pathway. It accepts NEDD8 from the E1 complex and catalyzes its covalent attachment to specific targets, most notably cullin-5 in complex with the E3 ligase RNF7/RBX2. This neddylation activity is essential for the assembly and activation of Cullin-RING ligase complexes, which regulate the ubiquitylation and proteasomal degradation of various substrates. UBE2F specifically promotes the degradation of the pro-apoptotic protein NOXA, thereby conferring oncogenic growth advantages and resistance to radiation and chemotherapy in cancer cells. Clinically, UBE2F is associated with Three M Syndrome 1, a rare genetic growth disorder. Additionally, its role in facilitating HIV-1 Vif-mediated degradation of APOBEC3G and its upregulation in lung cancer highlight its significance in viral pathogenesis and oncology, positioning it as a potential target for therapeutic intervention.
