TNF superfamily member 13b (TNFSF13B), also known as B-cell activating factor (BAFF), is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. It is encoded by the TNFSF13B gene located on chromosome 13q33.3. This protein acts as a potent B cell activator, playing a crucial role in the proliferation and differentiation of B cells. It is expressed in B cell lineage cells and interacts with receptors TNFRSF13B/TACI, TNFRSF17/BCMA, and TNFRSF13C/BAFFR. BAFF is involved in the stimulation of B- and T-cell function and the regulation of humoral immunity. Abnormal expression of BAFF has been associated with autoimmune diseases and certain types of cancers, such as chronic lymphocytic leukemia, B-cell lymphoma, and multiple myeloma. The protein exists in both membrane-bound and soluble forms, with the soluble form being generated through cleavage from the membrane-bound form.
