Tachykinin precursor 3 (TAC3) is a secreted neuropeptide belonging to the tachykinin family. Following proteolytic processing of the preproprotein, mature NKB functions as a critical neurotransmitter in both the central and peripheral nervous systems. It serves as a master regulator of mammalian reproduction, acting within the arcuate nucleus alongside kisspeptin and dynorphin to modulate GnRH secretion and gonadal function. Beyond its neuroendocrine roles, TAC3 is expressed in the placental syncytiotrophoblast, where it influences vascular responses. Clinically, mutations in TAC3 are a well-established cause of normosmic hypogonadotropic hypogonadism, a disorder characterized by impaired pubertal development and reproductive insufficiency. Additionally, altered TAC3 expression is associated with pregnancy-induced hypertension and pre-eclampsia, reflecting its diverse biological activities in neuronal excitation, vasodilation, and smooth muscle contraction.
