Synaptic vesicle glycoprotein 2A (SV2A) is a membrane protein present in synaptic vesicles that plays a crucial role in regulating neurotransmitter release and synaptic function. Its expression is reduced in temporal lobe epilepsy patients, and this reduction correlates with the degree of hippocampal sclerosis, suggesting a link between SV2A levels and epilepsy pathogenesis. In Alzheimer's disease (AD), SV2A mRNA and protein levels are significantly decreased in the hippocampus of AD patients compared to controls, and this reduction is consistent with the location of amyloid-beta (Aβ) deposition, implying that SV2A may interact with key AD pathogenic proteins like Aβ. Abrogation of SV2A promotes Aβ production, further supporting its involvement in AD pathogenesis.
