Stress-induced phosphoprotein 1 (STIP1), also known as Hsp70-Hsp90 organizing protein (Hop), is a co-chaperone that facilitates the interaction between heat shock proteins Hsp70 and Hsp90, playing a crucial role in protein folding and stabilization. STIP1 is characterized by three tetratricopeptide repeat (TPR) domains that mediate protein-protein interactions, as well as two aspartate-proline (DP) domains involved in client activation. Dysregulation of STIP1 has been implicated in various diseases, particularly in cancer, where it is often overexpressed and associated with enhanced cell proliferation and tumor progression. For example, STIP1 promotes oncogenic signaling pathways by stabilizing key proteins involved in cell growth and survival.
