Sorting nexin family member 21 (SNX21) is a cytosolic and endosome-associated protein belonging to the sorting nexin family. It is characterized by a conserved N-terminal phox (PX) domain that binds phosphoinositides, facilitating its association with endosomal membranes. SNX21 also contains C-terminal tetratricopeptide repeat (TPR) motifs that serve as scaffolds for protein-protein interactions. A key functional partner is the Huntington’s disease protein, huntingtin (HTT), which SNX21 recruits to endosomes to influence endosomal organization and cargo trafficking. As a member of the PX-associated B subfamily, SNX21 participates in endosomal sorting and the maintenance of protein homeostasis. While it lacks a BAR domain, its role in scaffolding endosomal components suggests it is a critical regulator of neuronal endosomal dynamics and intracellular protein trafficking.
