Sorting nexin 16 (SNX16) is a membrane-associated member of the sorting nexin family, characterized by a phosphoinositide-binding PX domain and a C-terminal coiled-coil region. It localizes primarily to early and late endosomes, where it mediates the binding to phosphatidylinositol-3-phosphate-rich membranes to regulate endosomal maturation and membrane tubulation. SNX16 is essential for the trafficking of proteins, such as the epidermal growth factor receptor (EGFR) and the tetraspanin CD81, from early endosomes toward lysosomal degradation. Beyond its role in vesicular transport and cholesterol trafficking, SNX16 influences cell migration and signaling. Clinically, SNX16 is associated with the progression of bladder and colorectal cancers, where it has been shown to activate c-Myc signaling by stabilizing eEF1A2, thereby promoting tumorigenesis.
