SLURP1 (Secreted Ly6/uPAR-related protein 1) is an immunomodulatory protein that plays important roles in skin development, inflammation regulation, and cancer suppression. It acts primarily by binding to nicotinic acetylcholine receptors, especially α7-nAChR, as well as other receptors like EGFR and PDGFRα. SLURP1 inhibits neutrophil adhesion and migration, suppresses pro-inflammatory cytokine production, and modulates keratinocyte differentiation. Mutations in the SLURP1 gene cause Mal de Meleda, a rare autosomal recessive skin disorder characterized by palmoplantar keratoderma. In cancer, SLURP1 demonstrates antiproliferative and anti-migration effects on lung adenocarcinoma cells by downregulating the PI3K/AKT/mTOR signaling pathway. Additionally, SLURP1 has been shown to prevent inflammation-induced proliferation in intestinal epithelial cells by modulating NF-κB expression and reversing LPS-induced inflammatory responses.
