SLIT and NTRK like family member 5 (SLITRK5) is a type I single-pass transmembrane protein predominantly expressed in the central nervous system. It is characterized by extracellular leucine-rich repeat domains and a cytoplasmic region containing tyrosine residues. SLITRK5 localizes to synapses where it acts as a negative regulator of neurite outgrowth and modulates axon-dendrite adhesion through interactions with type IIa receptor protein tyrosine phosphatases (RPTPs). These activities promote inhibitory synaptogenesis and influence dendritic morphology and circuit formation. In bone tissue, SLITRK5 is expressed in osteoblasts and functions as a negative regulatory co-receptor for Hedgehog signaling, thereby restraining bone formation. Clinically, SLITRK5 is linked to neuropsychiatric disorders; deficiency in mouse models results in obsessive-compulsive disorder (OCD)-like behaviors, and human genetic studies associate SLITRK5 variants with OCD and related phenotypes.
