Sirtuin 5 (SIRT5) is a protein encoded by the SIRT5 gene in humans and is a member of the sirtuin family of proteins, which are homologs to the yeast Sir2 protein. SIRT5 is primarily localized to the mitochondrion and exhibits enzymatic activities as a deacetylase, desuccinylase, and demalonylase, capable of removing acetyl, succinyl, and malonyl groups from the lysine residues of proteins. It plays a crucial role in regulating mitochondrial metabolism, particularly in the urea cycle by deacetylating and activating carbamoyl phosphate synthetase (CPS1), which is the rate-limiting step in liver mitochondria. Mice with SIRT5 deletion show elevated ammonia levels after fasting, while overexpression leads to increased CPS1 activity, suggesting SIRT5's role in regulating blood ammonia levels. Additionally, SIRT5 has been implicated in energy metabolism and has over 700 protein substrates, including those involved in mitochondrial and cytosolic functions.
