S100 calcium binding protein A10 (S100A10), also known as p11, is a member of the S100 family that uniquely exists in a constitutively active conformation, independent of calcium binding. It typically forms a stable heterotetrameric complex with annexin A2 at the inner leaflet of the plasma membrane. This complex regulates diverse cellular processes, including membrane trafficking, exocytosis, and the surface expression of ion channels such as TASK-1 and CFTR. S100A10 also facilitates local plasmin generation, which is critical for fibrinolysis and extracellular matrix remodeling. Clinically, S100A10 is frequently overexpressed in various cancers, where it promotes tumor invasion and metastasis. In the central nervous system, altered expression of p11 is strongly associated with the pathophysiology of mood disorders, particularly major depression.
