Pancreatic polypeptide (PPY) is a hormone secreted by PP cells in the endocrine pancreas. It is a 36-amino acid peptide that regulates pancreatic secretion activities, liver glycogen storage, and gastrointestinal secretion. PPY is synthesized and secreted by PP cells primarily located in the head of the pancreas and is part of a family of peptides that includes Peptide YY (PYY) and Neuropeptide Y (NPY). Its secretion is increased after a protein meal, fasting, exercise, and acute hypoglycemia, and is decreased by somatostatin and intravenous glucose. PPY has been implicated in conditions such as anorexia nervosa, where plasma levels are elevated, and in certain endocrine tumors, which can increase its secretion. The Y4 receptor, with high affinity for PPY, mediates its actions, primarily inhibiting pancreatic secretion by acting on receptors in the brain to inhibit vagal output to the pancreas.
