Plexin B3 (PLXNB3) is a transmembrane protein belonging to the plexin family that functions primarily as a high-affinity receptor for semaphorin 5A (SEMA5A), mediating key roles in axon guidance, invasive growth, and cell migration, as well as acting as a cell-cell adhesion mediator. It stimulates neurite outgrowth and mediates Ca2+/Mg2+-dependent cell aggregation, and in glioma cells, SEMA5A activation of PLXNB3 leads to cytoskeletal reorganization and inhibition of cell migration through ARHGDIA-mediated inactivation of RAC1. PLXNB3 is expressed in various tissues and has several alternatively spliced isoforms, reflecting its diverse functional roles. Disease associations include links to autism spectrum disorder and conjunctival nevus, and genetic studies have shown that PLXNB3 variants are associated with verbal performance and white matter volume in the human brain, suggesting a role in neurodevelopmental and cognitive phenotypes.
