01AB03413

Phospholipid transfer protein (PLTP) is a plasma protein that plays a critical role in lipid metabolism by mediating the transfer of phospholipids and free cholesterol from triglyceride-rich lipoproteins, such as low-density lipoproteins (LDL) and very-low-density lipoproteins (VLDL), to high-density lipoproteins (HDL). Structurally characterized by a distinctive banana-shaped fold similar to cholesteryl ester transfer protein (CETP), PLTP functions by penetrating lipoprotein surfaces to form ternary complexes that facilitate lipid remodeling and regulate HDL particle size. The protein interacts with apolipoprotein A1 and A2 to participate in reverse cholesterol transport pathways. Beyond its physiological role in maintaining lipid homeostasis, PLTP dysfunction is well-established in the pathogenesis of dyslipidemia and cardiovascular disease. Consequently, it serves as a significant therapeutic target for treating lipoprotein abnormalities and reducing the risk of atherosclerotic complications.
