Pleckstrin homology and RhoGEF domain containing G1 (PLEKHG1) is a guanine nucleotide exchange factor (GEF) that specifically activates the small GTPases RAC1 and CDC42. By facilitating the exchange of GDP for GTP, PLEKHG1 regulates cell morphology and actin cytoskeletal reorganization. The protein is activated through interaction with the Src family kinase FYN, which phosphorylates PLEKHG1 at multiple tyrosine residues, including Tyr-720 and Tyr-801, thereby enhancing its GEF activity and promoting Rho-dependent signaling. PLEKHG1 is primarily localized in the nucleoplasm and shares significant sequence identity with its paralog PLEKHG2. Clinically, genetic variations in PLEKHG1 are strongly associated with white matter hyperintensities and the small vessel subtype of ischemic stroke. Furthermore, its involvement in signal transduction pathways has linked it to the progression of prostate cancer and panic disorder.
