Phospholipase C gamma 1 (PLCG1) is a cytosolic enzyme that plays a pivotal role in intracellular signaling by coupling receptor tyrosine kinases, such as EGFR and PDGFR, to downstream cellular processes. Upon activation by phosphorylation, PLCG1 hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) to generate the second messengers inositol trisphosphate (IP3) and diacylglycerol (DAG). These messengers trigger calcium release and protein kinase C activation, regulating cell proliferation, survival, migration, and cytoskeletal remodeling. Structurally, PLCG1 contains SH2 and SH3 domains that mediate its recruitment and activation. Clinically, PLCG1 is a major oncogenic driver; recurrent activating mutations and gene amplifications lead to constitutive signaling in various malignancies. Its overexpression is frequently associated with increased tumor invasiveness and poor prognosis, particularly in lower-grade gliomas.
