Phosphofurin acidic cluster sorting protein 1 (PACS1) is a protein-coding gene involved in the localization of trans-Golgi network (TGN) membrane proteins and controls endosome-to-Golgi trafficking. PACS1 binds to phosphorylated acidic cluster motifs on client proteins such as furin and mannose-6-phosphate receptor, mediating their proper sorting and localization within the cell. It interacts with clathrin adaptor complexes like AP-1 and AP-3, playing a crucial role in intracellular trafficking and membrane protein localization. PACS1 also contributes to immune evasion by HIV-1, redirecting MHC-I molecules to the TGN to reduce their surface expression. Pathogenic variants in PACS1 cause multisystemic syndromes characterized by intellectual disability, developmental delays, and craniofacial abnormalities. The gene encodes a 963-amino acid protein essential for normal development, including lymphocyte function. The PACS1 protein contains functional domains such as a furin-binding region and an autoregulatory domain phosphorylated by casein kinase 2, regulating its interactions.
