Nibrin (NBN) is a critical protein involved in the cellular response to DNA double-strand breaks (DSBs), functioning as a key component of the MRN (MRE11-RAD50-NBN) complex, which is essential for DNA repair mechanisms. Nibrin plays a pivotal role in recognizing DSBs and facilitating their repair by recruiting other proteins, such as MRE11 and RAD50, to the sites of damage. Nibrin contains several functional domains, including FHA and BRCT domains, which enable it to interact with phosphorylated proteins involved in DNA damage signaling. Beyond its role in DNA repair, nibrin is also implicated in regulating cell cycle checkpoints and maintaining genomic stability. Mutations in the NBN can lead to Nijmegen breakage syndrome, characterized by immunodeficiency, growth retardation, and a heightened risk of cancer due to impaired DNA repair capabilities.
