N-ethylmaleimide sensitive factor, vesicle fusing ATPase (NSF) is a ubiquitously expressed cytosolic AAA+ ATPase that functions as a homohexamer to drive intracellular membrane trafficking. NSF plays a fundamental role in the vesicle fusion cycle by binding to SNARE complexes via soluble NSF attachment proteins (SNAPs). Utilizing energy from ATP hydrolysis, it mechanically disassembles post-fusion cis-SNARE complexes, thereby recycling individual SNARE proteins for subsequent rounds of fusion. This chaperone-like activity is essential for nearly all trafficking events, including synaptic vesicle exocytosis and the transport of AMPA receptors. Consequently, NSF is a critical determinant of neurotransmitter release and synaptic plasticity. Clinically, dysfunction in NSF-mediated SNARE disassembly is implicated in various neurological disorders characterized by defects in synaptic transmission and cognitive function.
