NOTCH regulated ankyrin repeat protein (NRARP) is a small protein containing two ankyrin repeats that functions as a downstream effector and negative feedback regulator of Notch signaling. It plays a role in various developmental processes and is involved in the regulation of cell proliferation, differentiation, and apoptosis. NRARP stabilizes LEF1 protein, a key transcription factor in the Wnt signaling pathway, by blocking its ubiquitination. It is also involved in angiogenesis, acting downstream of Notch at branch points to regulate vascular density. NRARP expression has been found to be upregulated in several types of cancer, including non-small cell lung cancer (NSCLC), thyroid cancer, and breast cancer. High NRARP expression has been associated with poor prognosis in NSCLC patients and may serve as a potential biomarker for cancer progression. Downregulation of NRARP has shown antitumor activities in thyroid cancer cells.
