Macrophage stimulating 1 (MST1) is a secreted plasma protein structurally related to hepatocyte growth factor. It is synthesized as a single-chain precursor and proteolytically cleaved into disulfide-linked alpha and beta chains to become biologically active. MST1 signals primarily through the RON (MST1R) receptor tyrosine kinase, regulating diverse cellular processes including macrophage activation, epithelial cell dissociation, and ciliary motility. In the nervous system, it acts as a neurotrophic factor for cranial motoneurons, supporting neuronal survival and limiting inflammatory damage. Clinically, MST1 is associated with immunodeficiency 14 and primary sclerosing cholangitis. Dysregulation of the MST1–RON signaling axis is also implicated in the progression of various cancers and chronic inflammatory responses, where it modulates the behavior of both epithelial cells and the innate immune system.
