Leukocyte receptor tyrosine kinase (LTK) is a receptor-type protein tyrosine kinase of 864 amino acids, structurally composed of a 423-amino acid extracellular domain, a transmembrane domain, and a 415-amino acid cytoplasmic kinase domain. It belongs to the ros/insulin receptor kinase family and primarily functions in signal transduction pathways regulating cell growth, differentiation, and the secretory pathway, particularly involving endoplasmic reticulum export sites and ER-to-Golgi transport. LTK is activated by ligands ALKAL1 and ALKAL2, leading to phosphorylation and activation of downstream pathways like MAPK and PI3 kinase. It is expressed in hematopoietic cells, pre-B lymphocytes, neurons, and the adult hippocampus, influencing neuronal development and neurite outgrowth. Pathologically, LTK has been implicated in autoimmune diseases like systemic lupus erythematosus (SLE), glioma susceptibility, and is often upregulated in acute myeloid leukemia. Its signaling role in disease contexts relates to aberrant kinase activity and mutations affecting cell survival and proliferation.
