Lymphocyte specific protein 1 (LSP1) is an intracellular, cytoskeletal-associated F-actin-binding phosphoprotein expressed primarily in hematopoietic cells and vascular endothelium. It features an N-terminal acidic region and a conserved C-terminal basic domain that facilitates direct binding to the sides of F-actin filaments. Functionally, LSP1 localizes to the actin cortex and specialized adhesion structures known as podosomes, where it regulates actomyosin contractility, leukocyte adhesion, migration speed, and mechanosensory protrusive forces. Through these mechanisms, LSP1 modulates the transendothelial migration of neutrophils and other immune cells. Clinically, LSP1 dysregulation is associated with neutrophil actin dysfunction and has been implicated in the progression of certain malignancies, including granular cell leiomyosarcoma, highlighting its role in inflammatory responses and cancer cell invasion.
