Leukocyte immunoglobulin like receptor B1 (LILRB1) is an inhibitory receptor expressed on various immune cells, including myeloid cells, NK cells, and subsets of T and B cells. It contains four extracellular immunoglobulin-like domains and intracellular immunoreceptor tyrosine-based inhibitory motifs (ITIMs). LILRB1 binds to MHC class I molecules, including HLA-G, and transduces negative signals that suppress immune responses and cytotoxicity. Upon activation, LILRB1 recruits phosphatases like SHP-1 and CSK, which dephosphorylate key kinases involved in immune cell activation. LILRB1 plays crucial roles in immune tolerance, regulating phagocytosis, and modulating NK cell and T cell functions. In pathological conditions, LILRB1 has been implicated in cancer progression, where its expression on NK cells correlates with decreased cytotoxicity in breast cancer. Additionally, some pathogens exploit LILRB1 to evade immune responses.
