Interleukin 9 receptor (IL9R) is a type I transmembrane cytokine receptor that mediates interleukin-9 (IL-9) signaling by forming a functional heterodimeric complex with the common gamma chain (IL2RG). Upon ligand binding, IL9R activates the JAK/STAT pathway, particularly in lymphoid and myeloid cells, to drive the proliferation and differentiation of T cells, B cells, and mast cells. The receptor contains conserved extracellular motifs, such as the WSXWS motif, which are essential for proper folding and high-affinity binding. Clinically, IL9R is strongly associated with asthma and Th2-biased allergic diseases. Additionally, autocrine IL-9/IL9R signaling loops are implicated in the pathogenesis of lymphoid malignancies, including Hodgkin lymphoma and thymic lymphomas.
