IKAROS family zinc finger 4 (IKZF4), also known as Eos, is a DNA-binding transcription factor that plays crucial roles in lymphocyte development and immune regulation. It contains zinc finger domains for DNA binding and protein interactions, recognizing the 5'-GGGAATRCC-3' Ikaros-binding sequence. IKZF4 functions as a transcriptional repressor, interacting with other proteins like SPI1, MITF, and FOXP3 to regulate gene expression. It is essential for the inhibitory function of regulatory T-cells (Tregs) and mediates FOXP3-dependent gene silencing by recruiting corepressors. IKZF4 is involved in B-cell differentiation, proliferation, and maturation, as well as in T-cell apoptosis regulation. While it has not been directly implicated in leukemogenesis, IKZF4 has been associated with autoimmune conditions such as alopecia areata and vitiligo. Additionally, it has been identified as a potential tumor suppressor in non-small cell lung cancer.
