IKAROS family zinc finger 3 (IKZF3), also known as Aiolos, is a hematopoietic-specific transcription factor belonging to the Ikaros family of zinc-finger proteins, which also includes Ikaros and Helios. Structurally, IKZF3 contains both N-terminal zinc finger domains for DNA binding and C-terminal zinc fingers for protein-protein interactions, enabling it to regulate gene expression through chromatin remodeling, direct DNA binding, and formation of transcriptional complexes. IKZF3 plays a critical role in the regulation of B lymphocyte proliferation and differentiation, and its function is mediated by the sequential formation of homodimers and heterodimers with other Ikaros family members. Multiple isoforms of IKZF3 exist due to alternative splicing, and the protein is predominantly expressed in lymphoid tissues, reflecting its specialized role in immune cell development and function. Disease associations include its implication in immune-related disorders and certain hematological malignancies, as dysregulation of IKZF3 can disrupt normal lymphocyte development and contribute to pathogenesis in these contexts.
