HECT, UBA and WWE domain containing E3 ubiquitin protein ligase 1 (HUWE1) is a large HECT-type E3 ubiquitin ligase that plays a central role in cellular stress responses and protein quality control. It features a massive α-solenoid ring scaffold with multiple interaction modules, including WWE and UBA domains, which allow it to target a broad range of substrates for proteasomal degradation. HUWE1 regulates over 40 substrates involved in the DNA damage response, transcription, apoptosis, and autophagy. Pathologically, germline and de novo variants in HUWE1 are linked to X-linked neurodevelopmental disorders. Additionally, its dysregulation is implicated in the progression of various cancers, positioning HUWE1 as an emerging target for anticancer drug development.
