Homeodomain interacting protein kinase 2 (HIPK2) is a nuclear serine/threonine kinase that plays a pivotal role in regulating cell proliferation, differentiation, and apoptosis. It features a conserved bilobal kinase domain and multiple regulatory regions, including a homeodomain-interacting domain and a speckle-retention signal for nuclear localization. HIPK2 functions as a critical sensor of DNA damage, where it phosphorylates the tumor suppressor p53 at Ser46 to induce apoptosis. It also interacts with and modulates various other transcription factors and co-regulators to control the cell cycle and stress responses. Clinically, HIPK2 is recognized as a potent tumor suppressor; however, its dysregulation or hyperactivity is frequently observed in various cancers and chronic fibrotic diseases.
