Grainyhead like transcription factor 1 (GRHL1) is a highly conserved member of the grainyhead family of transcription factors, essential for epithelial development and the maintenance of the skin barrier. It functions as either a homodimer or a heterodimer with GRHL2 or GRHL3, binding to specific consensus DNA sequences to regulate genes involved in epidermal differentiation and hair anchorage. A key downstream target of GRHL1 is the desmosomal cadherin DSG1, which is critical for the formation of intercellular junctions. Structurally, GRHL1 utilizes an immunoglobulin-like DNA-binding domain to interact with the major and minor grooves of its target promoters. While its primary physiological role is centered on barrier integrity, reduced expression of GRHL1 is associated with epidermal defects that phenocopy the loss of DSG1. It may also play a role in regulating glucose homeostasis and has been implicated in certain cancers where impaired target-site recognition disrupts normal epithelial growth.
