G-patch domain and KOW motifs (GPKOW) is a conserved, nuclear RNA-binding protein of approximately 55 kDa that contains one G-patch domain and two KOW motifs, both of which are implicated in nucleic acid and protein interactions essential for pre-mRNA splicing, particularly as a component of the minor spliceosome involved in U12-type intron processing. The G-patch domain, about 48-50 amino acids in length, is rich in conserved glycines and is thought to facilitate binding to single-stranded RNA and DNA, while the KOW motifs, each around 27 amino acids, form β strands that provide non-overlapping binding sites for nucleic acids and proteins, structurally resembling Tudor domains. GPKOW function is regulated by protein kinase A (PKA), with evidence suggesting that PKA-mediated phosphorylation modulates its RNA-binding activity and potentially its role in post-transcriptional mRNA processing. Upregulation of GPKOW has been observed in some disease contexts, and its essentiality in cancer cell lines suggests a possible role in tumorigenesis or cell survival pathways.
