Glycerol-3-phosphate dehydrogenase 2 (GPD2) is a flavoprotein dehydrogenase located on the inner mitochondrial membrane that serves as the mitochondrial component of the glycerol phosphate shuttle. It catalyzes the oxidation of glycerol-3-phosphate to dihydroxyacetone phosphate while simultaneously reducing ubiquinone, thereby linking cytosolic NADH metabolism to the mitochondrial respiratory chain. GPD2 is essential for triglyceride metabolism, glycerophospholipid biosynthesis, and maintaining mitochondrial redox balance. Clinically, GPD2 dysfunction is associated with type 2 diabetes mellitus, where altered energy metabolism in beta-cells contributes to disease pathogenesis. In diabetic kidney disease, GPD2 deficiency exacerbates podocyte injury. Additionally, GPD2-generated ubiquinol has been shown to suppress lipid peroxidation, providing a defense mechanism against ferroptosis in various cancer types.
