Gastrokine 1 (GKN1) is a 18-20 kDa stomach-specific protein highly expressed in normal gastric mucosa but downregulated or absent in gastric cancer. It contains a BRICHOS domain and plays a crucial role in maintaining gastric mucosal integrity and epithelial homeostasis. GKN1 exhibits mitogenic activity, interacts with other proteins, and has anti-amyloidogenic properties. It is secreted and co-localizes with actin in gastric epithelial cells. GKN1 expression is reduced in H. pylori-positive chronic gastritis and gastric cancer, suggesting its potential as a tumor suppressor. The protein's downregulation occurs throughout the progressive stages of neoplastic transformation, from chronic gastritis to gastric cancer. GKN1 overexpression in gastric cancer cell lines activates Fas receptor expression and increases apoptosis, while recombinant GKN1 treatment reduces proliferation in gastric cancer cells.
