GDNF family receptor alpha 2 (GFRA2) is a glycosylphosphatidylinositol (GPI)-anchored cell-surface co-receptor that primarily binds the neurotrophic factor neurturin (NRTN). It also exhibits affinity for glial cell line-derived neurotrophic factor (GDNF). Upon ligand binding, GFRA2 forms a signaling complex with the RET tyrosine kinase receptor, activating downstream pathways such as MAPK to support the survival and differentiation of sympathetic and parasympathetic neurons. GFRA2 is predominantly expressed in the nervous system and is essential for maintaining enteric and retinal neuronal health. Clinically, GFRA2 is considered a candidate gene for RET-associated neurocristopathies, with established associations to Hirschsprung disease 1 and retinal degeneration, where impaired neurotrophic support leads to significant neuronal dysfunction.
