FYN binding protein 1 (FYB1), also known as ADAP (Adhesion and degranulation-promoting adapter protein) and SLAP-130 (SLP-76-associated phosphoprotein), is a multifunctional protein crucial for immune cell signaling and function. It is expressed in T cells, monocytes, mast cells, macrophages, and NK cells, but not in B cells. FYB1 plays a pivotal role in post-activation T cell signaling, lymphocyte cytokine production, cell adhesion, and actin remodeling. The protein has various binding domains, including SH3 domains and a FPPP-motif that binds the ENA/VASP protein family, facilitating actin remodeling and cell adhesion processes. FYB1 is critical for the activation and proliferation of T-helper cells (CD4+) and cytotoxic T cells (CD8+), and its dysregulation has been implicated in thrombocytopenia.
