Fibronectin leucine rich transmembrane protein 2 (FLRT2) is a type I single-pass transmembrane protein that mediates cell-cell adhesion, migration, and axon guidance. It is characterized by an extracellular region containing leucine-rich repeats and a fibronectin type III domain, which facilitate both homophilic and heterophilic interactions. FLRT2 interacts with various partners, including latrophilins, UNC5 family receptors, and fibroblast growth factor receptors, to regulate cortical neuronal migration and vascular development. In the immune system, it promotes monocyte-to-macrophage differentiation and enhances macrophage phagocytosis via UNC5B–Akt/mTOR signaling. Clinically, FLRT2 acts as a context-dependent tumor suppressor. In breast cancer, its promoter is frequently hypermethylated, leading to downregulation; the loss of FLRT2 expression is associated with increased tumor cell proliferation, migration, and adhesion.
