Fc receptor like 3 (FCRL3) is a type I transmembrane glycoprotein encoded by the FCRL3 gene on chromosome 1, belonging to the immunoglobulin receptor superfamily. Structurally, FCRL3 features five extracellular immunoglobulin-like domains, a transmembrane region, and a cytoplasmic tail containing both immunoreceptor-tyrosine activation (ITAM) and inhibitory (ITIM) motifs, enabling dual regulatory signaling. Functionally, FCRL3 modulates immune responses by promoting TLR9-induced B-cell proliferation, activation, and survival, while inhibiting antibody production and plasma cell differentiation; it also suppresses B-cell receptor-mediated signaling through interactions with SH2 domain-containing phosphatases, reducing tyrosine phosphorylation and calcium mobilization. In regulatory T-cells, FCRL3 expression is linked to a memory phenotype and diminished suppression of effector T-cell proliferation. Clinically, FCRL3 is associated with increased susceptibility to autoimmune diseases such as rheumatoid arthritis, systemic lupus erythematosus, and autoimmune thyroid disease, with specific promoter polymorphisms enhancing disease risk by upregulating gene expression.
