Endothelial differentiation related factor 1 (EDF1) is a highly conserved transcriptional coactivator that plays a dual role in both the nucleus and the cytoplasm. In the nucleus, it acts as a bridge between the TATA-binding protein and various gene-specific transcription factors, enhancing the activity of nuclear receptors such as NR5A1, LXRA, and PPARG, as well as ATF1/2 and CREB1. In the cytoplasm of endothelial cells, EDF1 functions as a calmodulin-binding protein. By sequestering calmodulin, it negatively regulates nitric oxide synthase activity and modulates nitric oxide signaling pathways. Functionally, EDF1 is involved in regulating endothelial differentiation, lipid metabolism, and hormone-induced cardiomyocyte hypertrophy.
