Dynein axonemal intermediate chain 2 (DNAI2) is a critical protein component of the outer dynein arm in motile cilia and flagella. It belongs to the dynein intermediate chain family and is essential for the assembly and structural stability of the outer dynein arm, which powers microtubule-based motor activity and ciliary beating. DNAI2 is primarily expressed in the respiratory epithelium and sperm flagella, where it ensures proper ciliary motility and left-right patterning during development. Pathogenically, biallelic loss-of-function mutations in DNAI2 are a known cause of primary ciliary dyskinesia type 9 (PCD9). This condition is characterized by immotile or dyskinetic cilia, leading to chronic sinopulmonary infections, bronchiectasis, and situs inversus.
