Defective in cullin neddylation 1 domain containing 4 (DCUN1D4) is a nuclear protein that plays a central role in the post-translational modification of the cullin family of proteins. It functions as an E3-like factor that facilitates the transfer of NEDD8 from E2 enzymes to the C-terminal domains of various cullins, including CUL1, CUL2, CUL3, CUL4A/B, and CUL5. This neddylation process is essential for the activation of cullin-RING E3 ubiquitin ligases, which regulate global proteostasis, cell-cycle progression, and antigen processing. By modulating the turnover of key regulatory substrates, DCUN1D4 influences diverse cellular stress responses. Clinically, aberrant DCUN1D4 activity and dysregulated neddylation pathways are associated with the development of neoplasms, including optic nerve sheath meningiomas.
