Defective in cullin neddylation 1 domain containing 1 (DCUN1D1) is a cytosolic and nucleoplasmic adaptor that functions as an accessory E3 ligase within the NEDD8 neddylation pathway. It facilitates the activation of cullin-RING ubiquitin ligases by binding to cullin-RBX1 complexes, the NEDD8-conjugating enzyme UBE2M, and CAND1. This interaction promotes the nuclear translocation of these complexes and enhances cullin neddylation, thereby regulating protein ubiquitination and cell-cycle proteostasis. DCUN1D1 contains a C-terminal PONY domain essential for its E3 activity and an N-terminal UBA domain that serves as a negative feedback regulator. Clinically, DCUN1D1 is frequently amplified and overexpressed in squamous cell carcinomas of the head, neck, and lung. In these contexts, it acts as an oncogene that drives tumor proliferation, invasion, and overall aggressiveness.
