Cytotoxic and regulatory T cell molecule (CRTAM), also known as CD355, is a transmembrane protein of the immunoglobulin superfamily, comprising 393 amino acids with two extracellular immunoglobulin-like domains (IgV and IgC) and a cytoplasmic PDZ-binding motif. CRTAM is predominantly expressed on activated CD8+ T cells, natural killer T (NKT) cells, NK cells, and a subset of CD4+ T cells, where it functions as an adhesion molecule facilitating cell-cell interactions, particularly through binding to its ligand Necl2 on dendritic cells. This interaction is crucial for T cell activation and the production of cytokines such as IFNγ, IL-17, and IL-22, and it enhances the cytotoxic activity of NK cells against tumors. CRTAM has been implicated in several disease contexts, including inflammation, asthma, malignant mesothelioma, and certain cancers, where its role in promoting pro-inflammatory cytokine production and modulating immune responses may influence disease progression and immunopathology.
