Corticotropin releasing hormone (CRH) is a 41-amino-acid neuropeptide that serves as the primary physiological regulator of the hypothalamic-pituitary-adrenal (HPA) axis. Produced in the hypothalamus, it stimulates the release of adrenocorticotropic hormone (ACTH) from the anterior pituitary, which in turn triggers glucocorticoid production. CRH exerts its effects by binding to the G protein-coupled receptors CRHR1 and CRHR2, primarily activating the cAMP/PKA signaling pathway. Beyond its endocrine functions, CRH modulates immune responses and intestinal barrier integrity. Clinically, dysregulated CRH signaling and HPA axis overactivity are well-established factors in the pathogenesis of stress-related neuropsychiatric disorders, including major depression and anxiety, as well as various inflammatory and metabolic complications.
