Cleavage and polyadenylation specific factor 1 (CPSF1) is the largest subunit of the CPSF complex, a nuclear multisubunit assembly essential for the 3′ end processing of eukaryotic pre-mRNAs. CPSF1 recognizes the conserved AAUAAA polyadenylation signal near the polyadenylation site, facilitating site-specific cleavage and subsequent polyadenylation, which are critical steps in mRNA maturation and stability. The CPSF complex, which includes CPSF1, CPSF2, CPSF3, CPSF4, FIP1L1, Symplekin, and WDR33, coordinates with other factors to ensure accurate mRNA 3′ end formation, impacting gene expression regulation and cellular function. Beyond its fundamental role in RNA processing, altered CPSF1 expression has been linked to several diseases. Elevated CPSF1 promotes tumor cell proliferation and is associated with poor outcomes in cancers such as hepatocellular carcinoma, ovarian, head and neck, and prostate cancers. Additionally, CPSF1 mutations are implicated in early-onset high myopia and its dysregulation is observed in diabetic retinopathy.
